Information on genetic diversity and genetic relationships among genotypes of Brassica carinata is currently limited. The objectives of this study were to evaluate patterns and levels of genetic diversity in B. carinata based on amplified fragment length polymorphisms (AFLP) as compared with Brassica juncea and Brassica nigra, and to evaluate agronomic and seed quality data for plants grown in the field in western Canada. A total of 296 AFLP bands
were generated from four primer pair combinations and scored for presence/absence in 66, 20 and 7 accessions of B. carinata, B. juncea and B. nigra, respectively. B. carinata was less genetically diverse than the other two species. Differences in diversity were evident in the proportion of polymorphic
loci within each species: 23, 35 and 50% for B. carinata, B. nigra and B. juncea, respectively. Pair-wise similarity measures based on the Jaccard coefficient were highest among accessions of B. carinata and showed the narrowest range: 0.911 (0.810–0.981) compared to B. nigra: 0.569 (0.438–0.660) and B. juncea: 0.715 (0.345–0.951). AFLP-based genetic distance information can be used by plant breeders to select diverse genotypes.
AFLPs are also useful for fingerprinting cultivars and two primer pair combinations were sufficient to uniquely identify all
the accessions of B. carinata. More variation among accessions was identified in the agronomic trial than had previously been described in studies of B. carinata in western Canada, but the data were too limited to draw conclusions regarding specific accessions. Overall, the findings

Microbial biomass C and N, and activities related to C and N cycles, were compared in needle and leaf litter, and in the uppermost 10 cm of soil under the litter layer in Scots pine (Pinus sylvestris L.), Norway spruce (Picea abies L.) and silver birch (Betula pendula L.) stands, planted on originally similar field afforestation sites 23–24 years ago. The ground vegetation was differentiated under different tree species, consisting of grasses and herbs under birch and pine, and mosses or no vegetation with a thick layer of needles under spruce. The C:N ratio of the soils was 13–21 and the soil pHCaCl
2 3.8–5.2. Both showed little variation under different tree species. Microbial biomass C and N, C mineralization, net ammonification, reduction) did not differ significantly in soil under different tree species either. Birch leaf litter had a higher pHCaCl
2 (5.9) than spruce and pine needle litter (pH 5.0 and 4.8, respectively). The C:N ratio of spruce needles was 30, and was considerably higher in pine needles (69) and birch leaves (54). Birch leaves tended to have the highest microbial biomass C and C mineralization. Spruce needles appeared to have the highest microbial biomass N and net formation of mineral N, whereas formation of mineral N in pine needles and birch leaves was negligible. Summary Soil respiration was investigated in three loamy Orthic Luvisols (two arable, one forest soil), three sandy Haplic Podzols (also two arable, one forest soil) with a modified intersite method according to Lundegardh (1924). The method allows characterization of the CO2-flux from the soil and interpretation of the different levels with regard to temperature, nutrient and air supply. The method is sensitive to tillage and fertilization effects. In the two arable Luvisols the mean cumulative respiration rate was not uniform compared with the forest soil; in one case it was much higher and in another much lower. CO2 evolution in the Podzol under spruce was much lower than in the two arable Podzols. In the sandy Podzols 5 replicate measurements gave adequate results, with an error probability of 10%, but in the loamy Luvisols it was necessary to use 10 replicates to specify the same degree of difference. If soil respiration is very high, immediately after fertilization with cattle slurry or dung on arable land, or after litterfall in a deciduous forest, more replicates are necessary. 相似文献

Pigeonpea [Cajanus cajan (L.) Millsp.] is an important legume crop in South Asia, East and southern Africa, and the Caribbean. Pod fly (Melanagromyza obtusa Malloch) and pod wasp (Tanaostigmodes cajaninae La Salle) are important constraints to increase the production and productivity of pigeonpea under subsistence farming conditions. Host plant-resistance can be used as an important component for the management of these pests, and therefore, we evaluated 28 accessions of wild relatives of pigeonpea for resistance to these pests. There were significant inter- and intra-species differences in the relative susceptibility to pod fly and pod wasp damage. Accessions belonging to Cajanus scarabaeoides (L.) Thouars, C. sericeus (Benth. ex Bak.) van der Maesen, Rhynchosia bracteata Benth. ex Bak., C. acutifolius (F.v. Muell.) van der Maesen, C. lineatus (W. & A.) van der Maesen, and C. albicans (W. & A.) van der Maesen showed resistance to pod fly damage, while those from C. platycarpus (Benth.) van der Maesen, C. cajanifolius (Haines) van der Maesen and R. aurea DC. were susceptible. For the pod wasp, some of the accessions from C. scarabaeoides, C. albicans, Flemingia stricta Roxb., and R. bracteata (Roxb.) Wight showed a resistant reaction, while ICPW 83 belonging to C. scarabaeoides showed a susceptible reaction. ICPW 141, ICPW 278, and ICPW 280 (C. scarabaeoides), ICPW 214 (R. bracteata), ICPW 14 (C. albicans), and ICPW 202 (F. stricta) showed resistance to both pod fly and pod wasp damage. There was considerable variation in accessions belonging to different species for their susceptibility to pod fly and pod wasp, which can be exploited to breed for resistance to these pests. There was a negative association between pod wasp and pod borer damage, and therefore, it is important to keep track of the relative susceptibility of pigeonpea genotypes to pod wasp, while breeding for resistance to pod borers. 相似文献

The relative resistance of Manx Loaghtan, Shetland and Southdown sheep to naturally acquired gastrointestinal nematode infection was assessed over a ten week period. Thirty shearling ewes, ten from each of the three breeds, were co-grazed on an infective pasture. The animals were initially treated with an anthelmintic to minimise previous nematode infection, and faecal samples were collected from individual animals every two weeks. Southdown sheep had significantly higher faecal egg counts (FEC) than Shetland by week 6, and than Manx Loaghtan by week 10. After ten weeks there was an 86-fold difference in FEC between Shetland and Southdown sheep, and a 14-fold difference between Manx Loaghtan and Southdown sheep. These preliminary results suggest that primitive Shetland and Manx Loaghtan sheep may have greater resistance to gastrointestinal parasites, a trait that could be of value to the UK livestock industry. 相似文献

To optimize control of an avian influenza outbreak knowledge of within-flock transmission is needed. This study used field data to estimate the transmission rate parameter (β) and the influence of risk factors on within-flock transmission of highly pathogenic avian influenza (HPAI) H7N7 virus in the 2003 epidemic in The Netherlands. The estimation is based on back-calculation of daily mortality data to fit a susceptible-infectious-dead format, and these data were analysed with a generalized linear model. This back-calculation method took into account the uncertainty of the length of the latent period, the survival of an infection by some birds and the influence of farm characteristics. After analysing the fit of the different databases created by back-calculation, it could be concluded that an absence of the latency period provided the best fit. The transmission rate parameter (β) from these field data was estimated at 4.50 per infectious chicken per day (95% CI: 2.68–7.57), which was lower than what was reported from experimental data. In contrast to general belief, none of the studied risk factors (housing system, flock size, species, age of the birds in weeks and date of depopulation) had significant influence on the estimated β. 相似文献

Summary Denitrification activities were measured over a 3-year period in a coarse sandy soil and a sandy loam soil. In all years the crops were spring barley in combination with Italian ryegrass as a catch crop. The denitrification loss was measured using the acetylene inhibition technique on soil cores. Furthermore, a simple model was developed, based on daily values of soil moisture and soil temperature, to calculate the denitrification loss. Soil temperatures for the model were measured, whereas soil moisture was derived from a water-balance model. Measurements of denitrification gave an annual loss of 0.6 kg N ha-1, and the model calculated a loss of 1–2 kg N ha-1 in the coarse sandy soil. In the sandy loam soil annual losses were measured as 1.5, 3.0, and 13.0 kg N ha-1 in 1988, 1989, and 1990, respectively.
